Effect of Immunization Routes of Hepatitis B Surface Antigen on the Induction of Immune Responses in Mice

Abstract
To compare the mucosal immune response to hepatitis B surface antigen (HBsAg), mice were immunized with HBsAg intramuscularly, intranasally or intravaginally without adjuvant or with cholera toxin as a mucosal adjuvant. Vaccination routes affected the mucosal and systemic immune responses to HBsAg intravaginally administered into mice. Intramuscular injection of HBsAg resulted in the induction of strong systemic responses. Systemic immune responses following mucosal administration of HBsAg were generally weaker than following systemic administration. Salivary immune responses were higher following nasal administration than intramuscular or intravaginal immunization. HBsAg specific IgA was higher in vaginal washes following intravaginal than intranasal or intramuscular immunization. Intravaginal coadministration of HBsAg and cholera toxin showed higher IgA responses than intramuscular administration. Our results indicate that the vaccination route of HBsAg affects the induction of mucosal and systemic immune responses and that it may be important for vaccines for mucosal infection with HBsAg and for other viruses.
